First off, single-player can really help your movement. I find it very helpful to try to complete certain levels as fast as you can, and before you know it, you'll be moving around like a pro, hopping in and out of squid form, just popping out to splat enemies.
Another thing to be considered is how perfect single player is to practice your aiming, especially if you use gyro-scope controls. As you first start out with motion controls, your aim will probably be a little shaky (At least it was for me!) and it will give you a harder time splatting enemies. Single player gives you great stage layouts to practice jumping out and splatting enemies, splatting enemies while walking, etc. The best part is, if they manage to splat you first, you don't ruin your win streak or your rank.
